摘要
目的研究芹菜籽提取物(celery seed extracts,CSE)诱导卵巢癌SKOV-3细胞凋亡的作用。方法培养卵巢癌细胞株SKOV-3,配制不同浓度的芹菜籽提取液混合物(CSE)溶液,以2,2-二苯代苦味肼基自由基(DPPH)实验检测芹菜籽粗提物的抗氧化活性;应用激光共聚焦显微镜观察其凋亡的形态学表现,Western blot检测Caspase-3蛋白的表达。结果一定浓度范围的CSE对SKOV-3细胞的增殖均有抑制作用,且呈量效关系,激光共聚焦显微镜观察CSE作用12 h后SKOV-3细胞凋亡显著。Western blot分析表明药物处理后SKOV-3细胞的Caspase-3蛋白表达增加。结论 CSE能抑制卵巢癌SKOV-3细胞的增殖,促进其凋亡,其作用机制可能与提高Caspase-3活性有关。
Objective:To explore the the effect of celery seeds of extracts (CSE)on the apoptosis of ovarian cancer cells SKOV-3 and its related mechanism. Methods: 2, 2-2 benzene generation by the bitter taste and free radicals (DPPH) lab testing was used for antioxidant activity of CSE. The human ovarian cancer cell line SKOV-3 was treated with different concentrations(0.05、0.5、5?g/L) of CSE for 12h. The inhibition of cell proliferation and apoptosis were detected by morphological performance by Laser Scanning Confocal Microscope (CLSM) and the expression of Caspase-3 protein was measured by Western blot.Results: CSE had strong antioxidant activity and different concentrations of CSE had certain inhibitory effect on SKOV-3 cell proliferation, in a time-dependent manner.CSE induce cell line SKOV-3 apoptosis and the expressions of Caspase-3 protein in SKOV3 cells was up-regulated( P〈0. 05). Conclusion: CSE can inhibit the proliferation and induce the apoptosis of SKOV-3 cells,and its mechanism might be related to the effective of increasing the activity of Caspase-3 .
