β-桐酸诱导膀胱癌T24细胞凋亡机理 被引量:1

摘要 应用Hoechst33342染色对T24细胞进行染色,荧光显微镜下观察T24细胞经β-桐酸处理后的细胞形态学变化;应用流式细胞术分析β-桐酸对T24细胞凋亡率的影响,结果发现,β-桐酸能够诱导膀胱癌T24细胞凋亡。利用蛋白免疫印迹法分析凋亡的相关蛋白表达,发现β-桐酸能上调Bax/Bcl-2比例,并使Caspase-3表达水平上升,这为β-桐酸诱导T24细胞凋亡提供了进一步依据。β-桐酸还能够促进PPARγ的活化,且PPARγ抑制剂能够拮抗β-桐酸对JEG-3细胞生长的抑制作用和凋亡蛋白的表达影响,提示β-桐酸诱导膀胱癌T24细胞的凋亡作用与PPARγ的活化有关。 The morphological changes of T24 cell stained with hoechst 33342 staining assay was observed by fluorescence microscope and the ratio of cell apoptosis after incubation with β-eleostearic acid was determined by flow cytometry. β-eleostearic acid could inhibit the proliferation of T24 cells through apoptosis induction. In the western blot assay, Caspase-3 level and the ratio of Bax/Bcl-2 in T24 cells were increased along with β-ESA treatment. PPARγ was activated after β-ESA treatment, and the viability of T24 cells would rise if the expression of PPARγ, was blocked by its inhibitor. These suggested that β-eleostearic acid could induce T24 cell apoptosis through PPARγ, activation.
