乌萸汤含药血清对大鼠卵巢颗粒细胞P38MAPK信号转导途径的影响 被引量:3

摘要 目的:运用血清药理学方法观察乌萸汤含药血清对大鼠卵巢颗粒细胞P38丝裂原激活的蛋白激酶(MAPK)信号转导途径的影响。方法:25只SD大鼠随机分为正常对照组、西药对照组、乌萸汤低、中、高剂量组,给药4 d后采血并分离血清。然后在大鼠卵巢颗粒细胞培养体系中加入各组血清,培养48 h后,用Western blot测定颗粒细胞P38 MAPK磷酸化蛋白的相对表达。结果:与空白对照组比较,各给药组颗粒细胞P38 MAPK磷酸化蛋白相对表达量明显增多(P<0.01),乌萸汤中剂量组效果优于西药组(P<0.01)。结论:乌萸汤含药血清通过提高P38 MAPK蛋白磷酸化水平从而促进卵巢颗粒细胞增殖,表明乌萸汤是通过调控P38 MAPK信号转导途径以促进卵泡发育成熟及排出。 Objective:To research the effects of Wuyu decoction drug-containing serum on expression of P38 MAPK of ovarian granulosa cells (GC) of rats by serum pharmacological method. Methods:25 SD rats were randomly divided into normal con-trol group,western medicine group,low,middle and high dosage of Wuyu decoction group. Blood was taken from femoral artery of rats and serums were separated 4 d after administration. Serum in each group was added into ovarian granulose cell medium system of rats,then 48 h later relative expression of P38 MAPK phosphorylated protein was measured by Western blot. Results:Compared with blank control group,relative expression of P38 MAPK phosphorylated protein was significantly increased in treatment groups (P〈0.01). Effect of middle dosage group of Wuyu decoction was better than that of western medicine group (P〈0.01). Conclusion:Wuyu decoction drug-containing serum could promote the proliferation of ovarian granulosa cells through enhancing P38 MAPK protein phosphorylation. It is indicated that Wuyu decoction cou ld promote development,maturation and ovulation of ovarian follicle through adjusting P38 MAPK signal transduction pathway.
