蝉翼藤有机酸提取物抑制鸭体内乙型肝炎病毒复制及保肝作用的研究

The organic extract from the Securidaca inhibits duck hepatitis B virus replication in vivo and hepatoprotective effect

目的研究蝉翼藤有机酸类提取物(CYTYJS)对鸭乙型肝炎病毒(DHBV)复制的抑制作用与保肝的作用。方法将DHBV DNA阳性的广西麻鸭,随机分成5组,分别为CYTYJS高、中、低剂量组,模型组和拉米夫定(3TC)组。于用药前(T0)、用药7 d(T7)、14 d(T14)、21 d(T21)及停药后3 d(P3)分别采血,检测血清中DHBV DNA的滴度、丙氨酸氨基转移酶(ALT)和天门冬氨酸氨基转移酶(AST)的含量,实验结束时,取肝组织做病理学检查。结果与模型组比较,CYTYJS各剂量组用药后血清DHBV DNA滴度显著降低(P<0.05或P<0.01),停药3 d后,CYTYJS各剂量组DHBV DNA、ALT和AST仍在较低水平(P<0.05)而阳性组则有明显回升现象;镜下观察各组肝脏组织切片,与模型组比较CYTYJS高、中、低剂量组肝脏的病理改变有明显改善。结论 CYTYJS可能有一定的抗乙型肝炎病毒和保肝的作用。

Objective To study the effects of organic acid from the on anti-DHBV and protecting the liver activity. Methods the DHBV-DNA positive ducks from GuangXi Zhuang Autonomous Region were randomly divided into five groups：model group, positive group, CYTYJS-H, CYTYJS-M, CYTYJS-L group. The before and at the 7 th, 14 th, 21 th day during treatment, and at the 3 th day after finishing the treatment blood was collected to determined the serum content of DHBV-DNA, ALT, AST. The histopathological changes of liver were examined finally. Results compared to the model group, After treatment CYTYJS each group serum DHBV DNA titer decreased significantly （P＆lt;0.05 or P＆lt;0.01）. And at the 3 th day after withdrawal the DHBV DNA, ALT and AST level of CYTYJS each group still kept on lower level, showed certain inhibiting effect on the DHBV but the positive group rebounded quickly. The liver biopsy was observed under the light microscope, the pathological changes in the CYTYJS treatment group and the positive group had more improvement than that in model group. Conclusion CYTYJS maybe have some anti-hepatitis B virus and hepatoprotective effects.
