摘要
目的建立液相色谱-荧光检测法测定辣椒及花椒制品中罗丹明B。方法样品经甲醇/水/甲酸溶液(50+50+1)提取,Oasis MCX固相萃取小柱净化,Waters SymmtryC18柱(4.6 mm×250 mm,5μm),流速1.0 ml/min,梯度洗脱,柱温25℃,进样量50.0μl,激发波长538 nm,发射波长573 nm,外标法定量。结果罗丹明B在0.1 ng/ml^100.0 ng/ml范围均具有良好的线性(r>0.9999),回收率>89%,最低检出限为0.3μg/kg范围。结论该方法检测灵敏度高、特异性强、干扰少、检出限低、回收率好,可以用于辣椒和花椒制品中罗丹明B的检测。
Objective A high performance liquid chromatography -fluorescence detection method was developed for rhodamine B detection in capsicum and pepper products. Methods The sample was extracted with methanol/water/formic acid(50 + 50 + 1 ), and purified with Oasis MCX SPE cartridges, then separated by Waters Symmetry Cls column(4.6 mm x250 mm, 5μm) with gradient elution at a flow rate of 1.0 ml/min, excitation wavelength (λex ) of 556 nm and emission wavelength ( horn ) of 573 nm respectively, quantified by external standard method. And The column temperature was 25 ℃ and the injection volume was 50.0 ILl. Results Rhodamine B showed good linearity in the range of 0. 1 ng/ml - 100.0 ng/ml with eorrelation coeffi- cient greater than 0. 9999. The recoveries for rhodamine B were more than 89% , and the detection limit was 0.3 μg/kg. Conclusion Due to advantages of high sensitivity and specificity, less interference, low limit of deteetion and good recovery, this method can be applied to the determination of the rhodamine B in capsicum and pepper products.
