柴胡疏肝散对便秘模型大鼠便质及肠道蠕动功能的影响

Chaihu Shugan power impact on the quality of the stod and constipation rat model of intestinal peristalsis

目的通过观察柴胡疏肝散对便秘模型大鼠疗效,对便秘从肝论治进行探讨。方法 SD大鼠60只随机分为柴胡疏肝散组、模型组、空白对照组,每组20只(雌、雄各10只);除空白对照组外其余组大鼠用复方地芬诺酯片造便秘模型;造模成功后,柴胡疏肝散组给予柴胡疏肝散汤剂灌胃治疗,模型组给予等量蒸馏水灌胃,空白对照组不做处理,连续14 d;记录治疗最后7 d各组大鼠大便次数、大便重量。治疗结束后处死各组大鼠,测大鼠活性炭推进速率,血浆中胃动素、胃泌素、P物质浓度,远段结肠水通道蛋白3(AQP3)表达情况。结果柴胡疏肝散组与模型组比较,大鼠活性炭推进速率增加,大便总重量增加,AQP3表达上调,血浆中胃肠激素(胃动素、胃泌素、P物质)浓度上升。柴胡疏肝散组与空白对照组比较,各项指标均接近正常,但仍未达到正常水平。结论以柴胡疏肝散汤剂治疗便秘模型大鼠,其便秘症状得到改善,说明该方剂汤剂治疗便秘大鼠确有疗效。

Objective To observe the curative effect of Chaihu Shugan powder for rat model of constipation and to explore it from liver. Methods 60 SD rats were randomly divided into treatment group, vehicle group, and control group （20 rats in each group, 10 male and 10 female ones in each group）. The constipation model was induced by Compound Diphenoxylate Tablets. After the constipation model was induced successfully, the treatment group was administrated with decoction of Chaihu Shugan txnvder by gavage, and vehicle group was given distilled water with the same dose, and nothing to the control group. The treatment was lasted for 14 days. During the process, the defecation frequecy and stool weight of the last 7 days were recorded. All rats were killed at the end of the treatment, then the carbon advance rate was activated, content of plas- matic motilin, gastrin, and substance P and expression of AQP3 in distal colon were tested. Results Compared with vehicle group, the activated carbon advance rate and stool weight of treatment group were increased, expression of AQP3 was upregu- lated, and content of plasmatic motilin, gastrin, and substance P were increased. Compared with control group, all indexes of treatment group were close to normal level, but not exactly normal. Conclusions The constipation symptoms of rats＇ model were improved by treating with decoction of Chaihu Shugan powder, which indicates it is effective for constipation. There-fore, we think the fundamental principle of TCM treatment for constipation is soothing liver.
