胎盘FTO基因表达与出生体重的相关性研究

Study on the correlation between FTO gene expression in placenta and birth weight

摘要 目的:探讨胎盘FTO(Fat Mass and Obesity Associated,FTO)基因的表达量和孕产期相关因素与新生儿出生体重的关系。方法:采用自制调查表,收集温州医科大学附属育英儿童医院产妇及新生儿的相关信息,应用实时荧光定量PCR检测胎盘FTO基因的mRNA表达。结果:收集93名新生儿和产妇资料以及胎盘组织标本。单因素分析结果显示,胎盘FTO基因表达随孕周增加呈先升高后下降的趋势;不同性别间胎盘FTO表达量差异无统计学意义(t=0.074,P=0.148)。FTO表达量在不同孕前BMI和孕期体重增加量间差异无统计学意义(Z=0.639,P=0.726)、(F=1.476,P=0.688)。相关分析显示,出生体重和胎盘FTO表达量呈正相关,差异有统计学意义(r=0.314,P=0.002)。多元线性回归分析表明,胎盘FTO表达量和孕期体重增加共同影响出生体重,差异有统计学意义。结论:新生儿出生体重受母亲孕期体重增加、胎盘FTO基因表达的共同影响。 Objective: To explore the relationship between fat mass and obesity associated (FTO) gene expression level in placenta, maternal factors and neonatal birth weight. Methods: A selfdesigned questionnaire was used to collect related relevant information of pregnant women and their neonates from Yuying Children's Hospital of Wenzhou Medical University, real -time fluorescent quantitative PCR was used to detect Fro mRNA expression in placenta. Results: The data of 93 neonates and pregnant women and placental specimens were collected; the results of univariate analysis showed that placental FTO gene expression level increased with gestational week and then decreased. There was no statistically significant difference in placental FTO expression level between boys and girls ( t = 0. 074, P = 0. 148 ) ; There was no statistically significant difference in placental FTO expression level among pregnant women with different BMI values before pregnancy and different body weight gains during pregnancy ( Z = 0. 639, P = 0. 726), ( F = 1. 476, P = 0.688 ) ; the results of correlation analysis showed that there was a significantly positive correlation between birth weight and placental FrO expression level ( r = 0. 314, P = 0. 002) ; the results of multivariate linear regression analysis showed that both placental FTO expression level and body weight gain during pregnancy affect birth weight, there was significant difference. Conclusion: Neonatal birth weight can be affected by maternal body weight gain during pregnancy and placental FTO gene expression level.
