摘要
目的:探讨胰岛素样生长因子ⅡmRNA结合蛋白(IMP3)及P16在宫颈上皮内瘤变及浸润性癌中的表达及临床病理意义。方法;采用免疫组化S-P二步法及原位杂交方法检测20例正常宫颈上皮(Normal cervical epithelium,NCE)、35例宫颈上皮内瘤变(Cervical intraepithelial neoplasm,CIN)、50例宫颈鳞状细胞癌(Squamous cervical carcinoma,SCC)组织中的IMP3蛋白及P16蛋白的表达情况。结果:IMP3蛋白在NCE、CIN、SCC中的阳性表达率分别为0%、37.1%、90.0%,两两比较差异均有统计学意义(P<0.05),IMP3蛋白表达与宫颈病变程度呈正相关(r=0.537,P=0.000 0)。P16蛋白在NCE、CIN和SCC中的阳性表达率分别为0%、82.8%、92.0%。SCC中IMP3、P16阳性表达率均与组织学分化有关(P<0.05),IMP3蛋白与P16蛋白的表达呈正相关(r=0.393,P=0.005)。结论:IMP3与P16表达可为早期宫颈癌的病理诊断提供一定的实验依据,可以将其作为宫颈癌转移潜能的生物学标记物之一。
Objective: To explore the expressions and clinicopathological significances of IMP3 and p16 in cervical intraepithelial neoplasia (CIN) and cervical invasive carcinoma~ Methods: Immunohistochemical SP method and hybridization in situ were used to detect the expressions of IMP3 protein and p16 protein in 20 cases of normal cervical epithelium (NCE), 35 cases of CIN, 50 cases of squamous cervical carcinoma (SCC) . Results: The positive expression rates of IMP3 protein in NCE group, CIN group, and SCC group were 0%, 37. 1%, and 90. 0%, respectively, there was statistically significant difference among the three groups ( P 〈 0. 05 ) . The expression of IMP3 protein was positively correlated with the degree of cervical lesion ( r = 0. 537, P = 0. 000 0) . The positive expression rates of p16 protein in NCE group, CIN group, and SCC group were 0%, 82. 8% , and 92. 0%, respectively. In SCC group, the positive expression rates of IMP3 protein and p16 protein were correlated with histological differentiation (P 〈 0.05 ), the expression of IMP3 protein was positively corrolated with the expression of p16 protein (r = 0. 393, P = 0. 005 ) . Conclusion : IMP3 and p16 expressions may provide a certain experitnental evidence for pathological diagnosis of early cervical cancer, which can be used as potential biomarkers of metastasis of cervical cancer.
