摘要
目的研究扶正散结汤拆方对Lewis荷瘤小鼠免疫调节的机制,为其发挥临床效果提供科学依据。方法建立Lewis移植瘤模型,随机分为空白组、模型组、顺铂组、扶正组、化痰组、活血组、清解组、联合组,连续用药后处死小鼠,ELISA法检测小鼠外周血清IL-2、IL-10、IFN-γ变化。结果造模后各组脾脏指数、IL-2、IFN-γ血清含量降低,IL-10均有不同程度升高,小鼠免疫受到不同程度的抑制。扶正散结汤各配伍组分比较研究发现,配伍分组及联用组的脾脏系数降低程度均低于模型组。在防止IL-2下降方面,扶正组、活血组效果优于DDP组;在防止IFN-γ含量降低方面,联用组效果显著,提示全方组对IFN-γ作用最好,单独使用各配伍分组的效果稍差;在防止IL-10升高方面,联用组效果最好,其次为活血组。结论扶正散结汤正确配伍在调节免疫方面具有一定的协同效应而发挥抗肿瘤作用。
Objective To study the regulation mechanism in Lewis tumor-bearing mice treated with the individual combinations of Fuzheng Sanjie Decoction so as to provide the scientific evidences on the clinical efficacy.Methods The model of Lewis implanted tumor was set up.The blank group,model group, Cisplatin group,Fuzheng group,Huatan group,Huoxue group,Qingjie group and combined -therapy group were randomized.After continuous medication,the mice were sacrificed.ELISA was used to detect the chan-ges of peripheral serum IL-2,IL-1 0 and IFN-γin the mice.Results After modeling,the spleen indica-tors and the levels of IL-2 and IFN-γwere reduced and the level of IL-1 0 was increased to different ex-tents in each group.The immunity of mice was inhibited.It was found in the comparison among the groups, the spleen coefficients in the individual herbal combined groups and the combined-therapy were all lower than that in the model group.Concerning to the prevention of IL-2 reducing,the results in Fuzheng group and Huoxue group were better than that in DDP group.In term of the prevention of IFN-γreducing,the re-sult in the combined -therapy group was significant.It was suggested that the complete formula group a-chieved the best effect on IFN-γ.The effect was slightly inferior in the individual combination groups.For the prevention of IL-1 0 increasing,the effect was the best in the combined-therapy group and that was bet-ter in Huoxue group.Conclusion The correct combination of Fuzheng Sanjie Decoction achieves the coordi-native effect in immune regulation and anti-tumor to certain extent.
