摘要
供体牛超排后用富含X-精子的性控精液人工授精是生产奶牛体内性控胚胎的有效方法。本研究旨在探索冷冻X-精液不同输精方法对蒙贝利亚×荷斯坦F1体内性控胚胎生产的影响。选择10头蒙贝利亚×荷斯坦F1青年母牛作供体,按4 d 8次递减方法肌肉注射FSH超排处理,发情后分别在12、24 h和36 h各输精1次,每次输精剂量分别为2剂、2剂和1剂0.25 mL细管冷冻X-精液(实验Ⅰ组),或者分别在发情12 h和24 h各输精1次,相应的输精剂量为2剂0.25 mL细管冷冻X-精液(实验Ⅱ组)。配种后第7天(发情当天为0 d)常规方法采集胚胎。结果表明:实验Ⅰ组和实验Ⅱ组供体牛头均胚胎总数(10.0 vs 11.0)、头均可用胚胎数(6.2 vs 6.4)和头均不可用胚胎数(3.8 vs 4.6)差异不显著(P>0.05);虽然两实验组之间可用胚胎比例(62.0%vs 58.2%)和不可用胚胎比例(38.0%vs 41.8%)差异不显著,但实验Ⅰ组头均不受精卵数极显著低于实验Ⅱ组(P<0.01)。因此,供体母牛超排发情后适当增加人工授精配种次数可减少不受精卵的比例。
It is an efficient way to obtain sexed-embryo in vivo from dairy donors inseminated with sex-sorted semen after superovulation. This study was carried out to examine the effects of different way of artificial insemination (A. I. ) on the sexed embryo production efficiency of Montbeliarde xHolstein F1 donors superovulated and inseminated with female sex--sorted semen. 10 MontbeliardexHolstein F1 heifers were selected as donors and superovulated in the way of 8 times intramuscular injection (i. m. ) of FSH in decreasing dosage in 4 days. After onset of estrus (0 h), the donors were inseminated with Montbeliarde bull female sex--sorted semen at 12, 24 h and 36 h with 2 straws, 2 straws and 1 straw, respectively (Group I), or at 12 h, 24 h with 2 straws of sex--sorted semen each time (Group II). The embryos were collected at 168-170 h by non-surgical flushing. The results showed that the number of total embryos, transferable embryos and un-transferable embryos recovered per donor of Group Ⅰ (10.0, 6.2 and 3.8) were similar to those of Group II (11.0, 6.4 and 4.6)(P〉0.05). The number of unfertilized eggs per donor of Group Ⅰ was extremely significantly lower than that of Group Ⅱ (P〈0.01), although the percentage of transferable and un- transferable embryo in Group Ⅰ (62.0%, 38.0%) were similar to those of Group Ⅱ (58.2%, 41.8%) (P〉0.05). In conclusion, it could significantly reduce the unfertilized eggs that heifer donors received three times of A. I with sex- sorted semen than that of twice A. Ⅰ. after superovulation.
