冠心康对大鼠缺血再灌注损伤诱导凋亡心肌细胞结构形态学的影响 被引量:1

摘要 目的:观察中药复方冠心康对大鼠缺血再灌注损伤诱导凋亡心肌细胞结构形态学的影响。方法:40只SD雄性大鼠随机分为4组,假手术组、MIRI模型组、冠心康组、盐酸地尔硫卓组。分组给药处理7天,采用结扎大鼠心脏左冠状动脉前降支30分钟,再灌注2小时复制大鼠MIRI模型,造模成功后,在MIRI结扎线以下区域至心尖取材,通过常规苏木素-伊红(HE)染色,光镜200倍下用图形分析系统定量观察单位视野全部心肌细胞核平均光密度、平均黑度值、心肌细胞核的直径、心肌细胞的面积和周长、最大直径及最小直径的变化。结果:模型组大鼠心肌细胞结构形态学指标明显下降,与假手术组比较差异有统计学意义(P<0.01);冠心康组和盐酸地尔硫卓治疗组心肌细胞结构形态学明显改善,与模型组比较差异均有统计学意义(P<0.05或P<0.01)。结论:冠心康对大鼠MIRI心肌组织具有明显的保护作用,其作用机制与保护心肌细胞结构形态学有明显的关系。 Objective:To observe the effects of traditional Chinese medicine compound Guanxinkang on myocardial cell structure morphology changes of rats with ischemia reperfusion injury.Methods:40 SD male rats were randomly divided into 4 groups:the sham-operated group,the MIRI model group,the Guanxinkang group,and diltiazem hydrochloride group,each group was given corresponding meclicine for 7 days,Ligated the left anterior descending branch on rats' heart for 30 minutes,and after 2 hours' reperfusion to build MIRI model.After successful establishment of the model,the materials were drew from the following area of MIRI ligature to the cardiac apex,and colored by regular wood grain-eosin (HE) staining,and the average optical density and average blackness values,the diameter of the myocardial cell nucleus,the area and perimeter of myocardial cells,and its maximum diameter and minimum diameter variations were quantitatively observed with graphical analysis system under the light microscope 200 times.Results:To compared with the sham-operated group,myocardial cell structure morphology index of rats in MIRI model group decreased obviously,and the difference was statistically significant (P < 0.01) ;to compared with MIRI model group,the myocardial cell structure morphologies of rats in Guanxinkang group and diltiazem hydrochloride group improved obviously,the difference was statistically significant (P <0.05 or P < 0.01).Conclusions:Guanxinkang has an obviously protective effect on rats MIRI myocardial tissues,and its mechanism of action has an obvious relationship with protection of the myocardial cell morphology.
