华蟾素对荷瘤小鼠肝癌生长及HGF/c-Met信号通路的影响 被引量:11

Effects of Cinobufotalin on Tumor Growth of Hepatocarcinoma- bearing Mice and HGF /c- Met Signaling Pathway

摘要 目的:观察华蟾素对荷瘤小鼠肝癌生长的影响,并探讨华蟾素对HGF/c-Met信号通路的影响。方法:利用小鼠肝癌细胞实体瘤模型,设立3个处理组,即荷瘤模型组、5-Fu组和华蟾素组。分别在不同时间切除肿瘤,计量肿瘤的体积,进而观察华蟾素对小鼠肿瘤生长的影响。采用3种不同的处理方法分别处理肝癌细胞,通过Western blot检测细胞中HGF和c-Met蛋白的表达情况;ELISA检测荷瘤小鼠血清中HGF表达。结果:5-Fu组和华蟾素组小鼠肿瘤生长缓慢,肿瘤体积均明显低于荷瘤模型组(P<0.05);5-Fu和华蟾素处理的肝癌细胞HGF和c-Met蛋白表达水平均明显降低(P<0.05);与荷瘤模型组比较,5-Fu组和华蟾素组小鼠血清HGF的表达水平分别降低75%和65%(P<0.05)。结论:华蟾素可以抑制HGF/c-Met通路蛋白的表达,抑制荷瘤小鼠肝癌的生长。 Objective:To investigate the roles of Cinobufotalin on the tumor growth of hepatocarcinoma-bearing mice,and explore the effect of Cinobufotalin on HGF/c-Met signaling pathway.Methods:The mice with hepatocarcinoma were devided into three groups,the tumor model group,5-Fu treated group and Cinobufotalin treated group.The mice were sacrified in different time points after treatment,and tumor volume was measured.The hepatocarcinoma cells were cultured by 3kinds of methods,and Western blot was performed to detect the protein levels of HGF and c-Met,the serum HGF expression was detenmined by ELISA.Results:To compare with model group,tumor in mice with 5-Fu and Cinobufotalin treatment growed slowly,and the tumor volumes were smaller (P < 0.05),the expression of HGF and c-Met in hepatocarcinom cells cultured with 5-Fu and Cinobufotalin was lowered significantly,the expression of serum HGF in 5-Fu and cinobu fotalin groups respectively decreased 75% and 65%.Conclusion:Cinobufotalin can inhibit HGF and c-Met expression,inhibit tumor growth.
