摘要
目的:研究液体发酵猪苓菌丝体的化学成分.方法:3000L发酵罐发酵生产猪苓菌丝体,95%乙醇提取,硅胶、凝胶等柱色谱方法纯化化合物,红外、质谱、核磁共振等谱学技术鉴定化合物的结构.结果:从猪苓菌丝体中分离鉴定了12个化合物,分别是:麦角甾醇(1)、麦角甾-7,22-二烯-3,5,6-三醇(2)、麦角甾-7,22-二烯-3-酮(3)、猪苓酮A(4)、猪苓酮B(5)、α-羟基二十四烷酸乙酯(6)、N-(2′-羟基二十四烷酰)-1,3,4-三羟基-2-十八鞘氨(7)、腺苷(8)、烟酸(9)、琥珀酸(10)、尿嘧啶(11)和尿苷(12).结论:化合物1~12均为首次从猪苓菌丝体中分离获得.
Objective: To study the chemical constituents of mycelium of Polyporus umbellatus (Pers) Fries. by liquid fermentation. Methods: Liquid fermentation was performed in 3 000 L fermentor. Mycelium of P. umbellatus was extracted by 95% ethanol. Chemical constituents were isolated and purified using silica gel and Sephadex LH-20 column. IR, MS and NMR spectral analysis were used to identify their structures. Results: Twelve compounds were isolated from mycelium of P. umbellatus and identified as: ergosterol ( 1 ), ergosta-7,22-dien-3,5,5-triol ( 2 ), ergosta-7,22-dien-3-one ( 3 ), polyporusterone A ( 4 ), polyporusterone B ( 5 ), a-hydroxytetracosanoic ethyl ester ( 6 ), N-( 2'-hydroxytetracosanoyl ) -1,3,4- trihydroxyl-2-octodecanine (7), adenosine (8) , nicotinic acid ( 9 ), succinic acid ( 10 ) , uracil ( 11 ) , and uridine (12). Conclusion: Compounds 1-12 were isolated from mycelium of P. umbellatus for the first time.
