HER2基因扩增和蛋白表达与胃癌临床病理因素的相关性

The correlation of HER2 gene amplification and protein expression with clinical pathological factors of human gastric cancers

目的:探讨HER2基因扩增和蛋白表达与胃癌不同临床因素的相关性。方法:回顾性分析河南省肿瘤医院2012年5月至2013年10月收治的1 002例胃癌患者术后肿瘤组织病理标本,应用免疫组化技术(IHC法)和双色银染原位杂交技术(SISH法)检测HER2基因扩增和蛋白表达情况,比较不同临床特点的胃癌患者亚组间HER2表达阳性率的差异。结果:1 002例术后肿瘤组织病理标本中HER2阳性208例(20.8%)。HER2蛋白过表达与Lauren分型、病理分化程度和浸润深度有明显的相关性,差异均有统计学意义(P<0.05)。结论:在肠型胃癌、分化较好的胃癌、胃癌全层浸润时HER2阳性率明显提高。

Objective： To investigate the correlation of HER2 gene amplification and protein ex- pression with clinical factors of human gastric cancers. Methods ： 1 002 tumor specimens from the patients with gastric cancer in Henan Province Cancer Hospital from May 2012 to October 2013 were retrospectively analyzed. Immunohistochemical technique （IHC method） and double color silver staining in situ hybridization technique （SISH method） were used to detect the expression of HER2 gene amplification and protein expression. The positive rates of HER2 among the subgroups with different clinical characteristics were compared. Results： 208 cases （20.8%） was positive in 1 002 tumor specimens. Overexpression of HER2 protein was significantly correlated with Lauren typing, pathological differentiation and infiltration depth. The differences were statistically signifi- cant （P 〈0. 05）. Conclusion ： The positive rates of HER2 in the gastric cancers with intestinal type, higher differentiation, full thickness of infiltration were increased significantly.
