顺铂对人胃癌细胞系p53及其异构体△133p53蛋白表达的影响

Investigate the Effect and Clinical Significance of the Expression of P53 Dealt with Cisplatin in the Human Gastric Cancer Cell Lines MKN45

目的探讨顺铂对MKN45胃癌细胞系p53及其异构体△133p53蛋白表达的影响及临床意义。方法 PRMI1640血清培养的MKN45胃癌细胞系分为不同浓度梯度顺铂实验组及未加药物对照组。应用免疫细胞化学法检测p53蛋白表达情况。结果免疫细胞化学结果显示DDP组药物血清作用后,△133p53随着药物浓度增加,表达呈减少趋势,△133p53在胃癌组织中的表达呈负相关关系(P<0.01)。结论在顺铂诱导胃癌细胞系MKN45生长抑制的实验中,△133p53异构体蛋白表现出与野生型p53蛋白相反的趋势,并提示△133p53异构体蛋白本身可以作为肿瘤生物学行为的重要预测因子。

Objective To investigate the effect and clinical significance of the expression of p53 dealt with eisplatin in the human gastric cancer cell lines MKN45. Methods The human gastric cancer cell lines MKN45 was cul- tured by serum PRMI1640. The cells were divided into cisplatin group,which was separated by concentration gradient, and the control group. The expression~ effect of p53 were observed by immunocytochemistry. Results Anti-oncogene p53 location in cytoplasm mainly was obselwed by immunocytochemistry. In DDP group,the expression of p53 reinforced obviously,and there was a positive correlation between cisplatin concentration and the expression strength of p53. There was an ohvious difference of the positive expression rate between control and DDP groups（ P 〈 0.01 ）. Conclusion The expression of p53 ,deah with the chemotherapeutics cisplatin, reinforce in the human gastric cancer cell lines MKN45. There is an important significance for studying the occurrence and development of gastric cancer.
