摘要
目的观察两种造模方法对大鼠慢性肾衰竭模型肾脏损伤的影响,为不同类型慢性肾衰竭动物模型的选择建立提供参考。方法两种方法建立慢性肾衰竭大鼠模型:(1)大鼠5/6肾切除(A组);(2)单侧肾切除联合尾静脉重复注射阿霉素(B组)。分别测定各组大鼠肾功能(尿素氮、肌酐)、血清白蛋白、C反应蛋白(CRP)、丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性,光镜下观察各组大鼠肾脏病理改变,并与正常大鼠(N组)作比较。结果 A组和B组大鼠均有血生化指标和肾脏病理改变,4、8、12周末时,两组大鼠血肌酐、尿素氮、CRP、MDA与N组相比显著增高(P<0.05,P<0.01),白蛋白、SOD与N组相比有显著降低(P<0.05);8周末时A、B组间MDA、SOD差异有统计学意义(P<0.05)。光镜下A、B组大鼠均有肾脏病理改变,肾小球及肾小管、肾间质均有损伤。8周末时,B组较A组肾间质炎症病理改变的程度和范围更加明显。结论单侧肾切除联合尾静脉重复注射阿霉素诱导的慢性肾衰竭模型与5/6肾切除诱导的慢性肾衰竭模型相比,微炎症状态及氧化应激失衡出现更早,肾间质更早出现炎症病理改变。
Objective To investigate the differences between two different methods in construction of rat nephropa-thy model, thus to provide references for building the different renal failure models. Methods The rat nephropathy mod-els were established by resection of 5/6 of the kidney (Group A) and by unilateral nephrectomy combined repeated injec-ting adriamycin (Group B). The renal function indexes (BUN and Cr), serum albumin, C-reactive protein (CRP), malondialdehyde (MDA) and superoxide dismutase (SOD) were recorded, and pathological changes were observed for comparison. All of above of the two groups were compared with normal rats (Group N). Results All blood biochemical indices of the experimental animals in Group A and B were changed after modeling. At the 4^th , 8^th and 12^th weekends, the concentrations of BUN, Cr, CRP and MDA in both Group A and B were significandy increased compared with Group N (P 〈 0. 05 ), while serum albumin and SOD were significantly reduced (P 〈 0.05 ). Moreover, the concentration of CRP and MDA in Group B were increased significantly more prominent than Group A at the 8th weekend (P 〈 0.01 ), while the reduction of SOD was also significantly more prominent (P 〈 0. 05). Focal renal interstitial inflammation was revealed in both Group A and B, while it was significantly severer in Group B at the 8^th weekend. Conclusion Earlier microinflam-matory state, oxidative stress state and renal interstitial inflammation are founded in the unilateral nephrectomy combined repeated injecting adriamycin model than in 5/6 of kidney resection model.
