TNF-α、IL-1在佐剂性关节炎大鼠血清中的变化

摘要 目的:观察佐剂性关节炎大鼠肿瘤坏死因子α( TNF-α)、白细胞介素(IL)-1β、一氧化氮( NO)、IL-18的变化,以探讨类风湿性关节炎( RA)的发病机制。方法将Wistar大鼠分成2组:正常组和模型组;模型组足部皮下注射弗氏完全佐剂,制造大鼠佐剂性关节炎模型( AA模型),对照组足部皮下注射生理盐水作对照;通过关节炎评分、X线评分、病理评分等评价炎性程度;免疫后第42天处死大鼠,取血后用ELISA法测TNF-α、IL-1β、NO、IL-18水平;取大鼠踝关节做苏木精-伊红染色。结果与正常组相比模型组血清TNF-α、IL-1β、NO水平明显升高(P <0.05)2,组IL -18水平差异无统计学意义(P>0.05)。结论佐剂性关节炎大鼠造模成功;模型组血清中TNF-α、IL-1β、NO含量较正常组明显升高,提示TNF-α、IL-1、NO可能参与了RA的发病及发展。 Objective To observe the changes of tumor necrosis factor -α( TNF-α) , interleukin-1 β( IL-1β), nitric oxide (NO) and interleukin-18 (IL-18) in rats with adjuvant-induced arthritis and to explore its mecha-nism of action .Methods Wistar rats were randomly divided into a control group and a model group .Rats in the model group was subcutaneously injected with the Freund′s complete adjuvant to establish a model of adjuvant -induced arthritis (AA model).Those in the control group were subcutaneously injected with normal saline alone .The severity of joint in-flammation was assessed according to arthritis index , radiographic changes and pathological grade .The rats were sacri-ficed 42 days later to examine the levels of TNF -α, IL-1β, NO, IL-18 by ELISA.The right stifle-joint of rats were taken for HE staining .Results Compared with the control group , the levels of serum TNF-α, IL-1βand NO in the model group were remarkably incrased (P〈0.05).There was no significant difference between the two groups as to the level of IL-18 (P〉0.05).Conclusion We have successfully established a rat model of adjuvant -induced arthritis. The levels of serum TNF-α, IL-1β, and NO in the model group are significantly higher than those in the control , indi-cating the potential role of these molecules in the pathogenesis of rheumatoid arthritis .
