摘要
为了研究Ghrelin是否参与影响体外受精(IVF)过程,试验运用体外成熟及体外受精技术,向体外受精培养体系中外源性地添加Ghrelin(0μg/L、300μg/L、600μg/L、900μg/L、1 200μg/L)及已被多项研究作为GHS-R的颉颃剂D-Lys3-GHRP-6(0μg/L、10μg/L、100μg/L、1 000μg/L)的方法,对Ghrelin及其受体颉颃剂D-Lys3-GHRP-6对绵羊卵母细胞体外受精情况进行检测,初步确定Ghrelin在绵羊体外受精过程中可能扮演的角色。结果表明:外源性添加的Ghrelin在绵羊卵母细胞体外受精过程中并没有发挥明显的生理作用,说明Ghrelin能促进卵母细胞的体外成熟而不能促进体外受精。
To study whether Ghrelin was involved in the process of in vitro fertilization( IVF), the effects of Ghrelin and GHS - R antagonist (D- Lys3 -GHRP- 6 )on IVF of sheep oocytes were detected by exogenous additions of Ghrelin (0 μg/L, 300 μg/L ,600 μg/L,900 μg/L, 1 200 μg/L)and D- Lys3 -GHRP- 6(0 μg,/L, 10 μg/L, 100μg/L, 1 000 μg,/L)to the IVF culture system, using the techniques for in vitro maturation and fertilization. It was initially determined that Ghrelin might play a role in the process of in vitro fertilization of sheep oocytes. The results showed that exogenous Ghrelin did not play an obvious physiological effect on the process of in vitro fertilization of sheep oocytes. The re- sults indicate that Ghrelin has a promotion effect to the in vitro maturation of sheep oocytes, but can not promote the in vitro fertilization.
