摘要
目的研究氟中毒大鼠肝组织中caspase-3、caspase-9 mRNA表达的变化。方法将48只健康清洁级SD雄性大鼠随机分为4组,分别为对照(去离子水)组和低(50 mg/L)、中(100 mg/L)、高剂量(200 mg/L)氟染毒组,每组12只。采用自由饮水方式进行染毒,连续染毒120 d。采用荧光定量PCR法检测肝组织中caspase-3、caspase-9 mRNA的表达水平。结果与对照组相比,高剂量氟染毒组caspase-3、caspase-9 mRNA的表达水平明显升高,差异均有统计学意义(P<0.05)。结论过量氟从基因水平促进大鼠肝脏组织中caspase-9、caspase-3的表达。
Objective To know the expression changes of caspase-3,caspase-9 mRNA in the liver tissue of rat with fluorosis. Methods A total of 48 cleanly Sprague-Dawley male rats were randomly divided into four groups (control,low-fluoride, moderate-fluoride and high-fluoride,12 in each group) and administered 0,50,100,and 200 mg/L of sodium fluoride, respectively,through drinking water for 120 consecutive days. caspase-9 and caspase-3 mRNA expression levels were analyzed by using real-time PCR. Results The fluoride treatment increased caspase-9, caspase-3 mRNA expression levels significantly (P〈0.05). Conclusion Excessive fluoride can promote the expression of caspase-9, caspase-3 mRNA in the livers tissue of rats.
