甘草药材HPLC指纹图谱研究 被引量:8

摘要 目曲:建立甘草药材HPLC指纹图谱,并尝试将其应用到质量评价中。方法:采用RP—HPLC(DAD)法,用AgilentExtend—C18色谱柱(250mm×4.6mm,5μm)和Extend-C18保护柱(20mm×4.6mm,5μm)。以乙腈(A)-0.026%磷酸水溶液(B)为流动相。采用梯度洗脱:0~20min,A为20%~25%;20—30min,A为25%~33%;30~55min,A为33%~50%;55—65min,A为50%-60%;65~80min,A为60%。检测波长为280nm。结果:对29批甘草药材进行测定,标定了29个共有峰,且样品基本上被分成了三类。结论:建立了高效液相指纹图谱(RP—HPLC),能有效地体现出中药成分的复杂性;对29批甘草药材进行的相似度评价,反应出一定的差异性,推测这三类甘草的差异可能为种的不同所致。 Objective: To construct HPLC Fingerprint Spectrum of Radix Glycyrrhizae and apply it into quality assessment. Methods: By the method of HPLC-DAD, the analysis was performed on an Agilent Extend-C18 column and Extend-C18. The mobile phase was consisted of acetonitrile (A) and aqueous H3 PO4 (B). The gradient elution of 20% - 25 % A at 0-20 rain ; 25%-33% A at 20-30 min; 33%-50% A at 30-55 min; 50%-60% A at 55-65 min; 60% A at 65-80 min. We detected the wave length was 280 nm. Results: Twenty-nine batches of Radix Glycyrrhizae were determined and 29 common peaks were demonstrated. The samples were clarified into 3 categories. Conclusion: The RP-HPLC was established to reflect the complexity of material medica; after evaluating the similarity of 29 batches of Radix Glycyrrhizae, we found there was significant difference among them, thus we speculate about these 3 categories of Radix Glycyrrhizae are different species.
