蛹虫草多糖对鸡脾脏淋巴细胞的影响 被引量:6

Effects of Cordyceps militaris Polysaccharides on Splenic Lymphocytes in Chicken

摘要 试验将7种浓度蛹虫草多糖(CMP)单独或与刀豆蛋白A(ConA)、脂多糖(LPS)同时加入鸡脾脏淋巴细胞的培养体系中,培养48 h时用MTT法测定淋巴细胞增殖(A570值)的变化;将5种浓度的CMP 40%乙醇提取物(CMP40)、CMP 50%乙醇提取物(CMP50)分别与ConA加入到鸡脾淋巴细胞的培养体系中,培养24 h后收集细胞培养上清液,测定白细胞介素-2(IL-2)和γ-干扰素(IFN-γ的含量.结果显示,CMP在31.25~2 000 μg/mL时,CMP40+ConA组的A570值显著高于ConA对照组;CMP在31.25~500μg,/mL时,CMP50+ConA组A570值显著高于ConA对照组,CMP40+LPS组、CMP50+LPS组的A570值显著高于LPS对照组;CMP40在125~1 000μg/mL时、CMP50在500 μg/mL时,各多糖组的A570值显著高于细胞对照组;CMP40和CMP50在250~1 000 μg/mL时,各多糖组的IL-2和IFN-γ浓度显著高于对照组.结果表明,CMP40和CMP50在合适的剂量能单独或协同ConA、LPS刺激鸡的T、B淋巴细胞增殖,促进淋巴细胞分泌IL-2和IFN-γ,从而增强鸡的细胞免疫能力. In this study, two kinds of Cordyceps militaris polysaccharides (CMP) at seven concentrations alone or with concanavalin A (ConA) ,lipopohysaccharide (LPS) were added to the culture system of chicken spleen lymphocytes, cultured for 48 h with MIT method for the determination of lymphocyte proliferation (A570)changes. Five concentrations of CMP extract by 40% ethanol(CMP40) ,CMP extract by 50% ethanol(CMP,o)respectively with ConA were added to the culture system of mouse spleen lymphocytes, and cell culture supernatant was harvested after 24 h for determination of concentrations of interleukin-2 (IL-2) and interferon-γ (IFN-γ). The results showed that CMP at 31.25 to 1000μg/mL, A570 values of CMP40+ConA and CMP,o+ConA group were significantly higher than those at ConA control group; CMP at 31.25 to 500 μg/mL, A570 values of CMP40+ LPS and CMP50+ LPS group were significantly higher than that at LPS group; CMP40 at 125 to 1 000 μg/mL group and CMP,o at 500 J..lgfmL group, A570 values was higher than that of cell control group. CMP40 and CMP,o at 250 to 1 000 μg/mL group, the concentrations of IL-2 and IFN-γ, were significantly higher than that of control group. The results indicated that CMP40 and CMP40 at suitable doses could alone or with ConA,LPS stimulate T,B lymphocyte proliferation of chicken,and promote lymphocyte secretion of IL-2 and IFN-γ thereby enhancing cellular immunity of chicken in vitro.
