摘要 [目的]研究柠檬[Citrus limon(L.)Burm.f.]种子萌发的适宜处理方法。[方法]以不同种皮剥离程度的柠檬种子(不剥种皮、只剥外种皮、剥除内外种皮)为试材,应用不同消毒剂[4%(V/V)蓝月亮、0.1%升汞]消毒不同时间(7、15、23 min),进行柠檬播种试验。[结果]0.1%升汞消毒8 min的效果优于4%(V/V)蓝月亮消毒15 min,但差异性不大;种子在剥除内、外种皮后萌发率、生长势情况都好于只剥外种皮和未剥除种皮的。[结论]考虑到升汞具有致癌变、致突变性等危害,在试验操作中不提倡使用,故以4%(V/V)蓝月亮处理15 min为柠檬种子消毒的最佳方式。该研究可为柠檬组织培养及柑橘无病毒苗木培育提供参考依据。 [Objective] The aim was to study the suitable treatment method of lemon seeds germination.[Method] Lemon seeds of without stripping the testa,only peeling the episperm,and peeling endotesta and episperm were sterilized by 4% (V/V) blue moon and 0.1% HgCl2 for 7,15,23 respectively,and then conducted on its sowing test.[Result] The disinfection effect of 0.1% HgCl2 for 8 min was better than that of 4% (V/V) blue moon for 15 min,but they had no significant difference.The germination rate and growth potential of lemon of peeling endotesta and episperm were superior to those of only peeling the episperm.[Conclusion] HgCl2 dose not advocate using in test process because of its damage of sending canceration and mutagenicity,so the best treatment method of lemon seeds is 4% (V/V) blue moon for 15 min.The study provides a reference basis for tissue culture of lemon and breeding of no virus seedlings of orange.
