摘要
目的:研究参蛤散对压力负荷性心力衰竭大鼠心肌组织NADPH氧化酶2、4表达的影响,并与法舒地尔进行比较。方法:腹主动脉缩窄建立大鼠压力负荷性心力衰竭模型,造模8周后,48只大鼠分为假手术组、模型组、参蛤散组和法舒地尔组。参蛤散组予参蛤散灌胃(1.89?g/kg),法舒地尔组予法舒地尔腹腔注射(5?mg/kg),假手术组和模型组以蒸馏水2?mL灌胃,每日均1次。16周后免疫组化和Western-blot方法检测心脏NOX2、4表达。结果:与假手术组比较,模型组NADPH氧化酶2、4的表达增高(P<0.01);与模型组比较,参蛤散组和法舒地尔组表达量降低(P<0.01),且参蛤散组优于法舒地尔组(P<0.01)。结论:参蛤散能抑制压力负荷性心力衰竭大鼠心肌组织NADPH氧化酶2、4的表达,作用优于法舒地尔。
Objectives : To explore the effect of Shen'ge powder on the expressions of NADPH oxidase 2,4 in the eardiomyoeyte in pressure-overload heart failure rats. Methods : Model of pressure-overload heart failure was established through incompletely constriction of abdominal aorta. After 8 weeks of the experiment,48 rats were divided into 4 groups : sham group, control group, fasudil group ( 5 mg/kg ) and Shen'ge powder group ( 1.89 g/kg, orally administrated ). 16 weeks laler, the rats were sacrificed to detect the expressions of NADPH oxidase 2,4 in the cardiomyocytes by the method of immunohistochemistry and Western-blot. Results : Compared with sham group, the expressions of NADPH oxidase 2,4 in control group were increased (P〈0.01). Compared with control group, the expressions of NADPH oxidase 2,4 were reduced in fasudil group and Shen'ge powder group ( P〈0.01 ), and /he latter was lower ( P〈0.01 ). Conelusion:Shen'ge powder could inhibit the expressions of NADPH oxidase 2,4 in the cardiomyocyte in pressure-overload heart failure rats, which is superior to the fasudil.
