摘要
【目的】检测CD4+/CD8+T细胞在公鸡生殖器官中的分布密度。【方法】通过荧光定量PCR分析IL-8的表达水平,以及通过冰冻切片和免疫组化技术,定位健康公鸡的生殖系统中CD4+和CD8+T细胞。【结果】IL-8在附睾中表达水平最高,在睾丸中的表达水平最低;有少量的CD4+和CD8+T细胞被定位在睾丸间质细胞区。在附睾管和输精管的固有层和黏膜上皮中,均识别到两种类型的T细胞。而且CD4+和CD8+T细胞在公鸡生殖道中不同部位密度的变化趋势明显:CD4+和CD8+T细胞在公鸡生殖管道中段(输出管、附睾管)的密度高于两端(睾丸网、输精管)。【结论】T细胞介导的免疫防御主要发生在附睾,尤其附睾输出管和附睾管中。
[Objective] The aim of this study was to localize the distribution of CD4 and CD8 positive T cell in reproductive organs of rooster. [Method] The expression level of IL-8 was examined by qRT-PCR, and CD4+-/CD8 + T cells were localized by immunohistochemistry on tyro-sections. [Results] IL-8 had a high expression in epididymis, and had a low expression in testis. Moreover, a few of CD4q-/CD8+ T cells were localized in the region of interstitial cell, and CD4 q-/CD8ff- T cells were localized in mucosal epithelium and lamina propria of the epididymal duct and duct defense. Moreover, the density change of CD4q-/CD8q- T cells in different segments of reproductive organs have significant difference, the density of CD4+/CD8+ T cells in middle segment (efferent duct and epididymal duct) was significantly higher than that the two terminals (rete testis, and duct deferens). [Conclusion] The results suggested epididymis, especially efferent duct and epididymal duet were the main segment of immunodefense by cell-mediated immunity.
