摘要
香蕉穿孔线虫是一种重要的植物寄生线虫,通过对香蕉穿孔线虫ITS区域序列分析,利用在线软件Primer Explorer V4设计了环介导等温扩增的内、外引物各1对(2条),环引物1对。通过对反应条件的优化,建立了香蕉穿孔线虫环介导等温扩增体系。实验结果表明该方法准确、特异、简便、快速。
Radopholus similis is one important plant parasitic nematode species. A specific loop-mediated isothermal amplification (LAMP) primers group, including Rs-3F3, Rs-3B3,Rs-3FIP, Rs-3BIP, Rs-3LF and Rs-3LB for R. similis were designed individually, through alignment and anylysis of internal transcribed spacer (ITS) and 5.8 S ribosomal DNA regions of Radopholus nematodes, using the online biological software Primer Explorer V4 Here, we present a LAMP detection method for R. similis, through temperature optimization. Our experimental results suggest that LAMP detection was precise, specific, simple and rapid.
