取食不同寄主植物对绿盲蝽热激蛋白AlHSP90表达的影响 被引量:2

Expression Analysis of Heat Shock Protein 90 Gene AlHSP90 in Apolygus lucorum(Meyer-Dür) after being Fed on Different Host Plants

摘要 绿盲蝽热激蛋白90基因AlHSP90是绿盲蝽体内重要的抗逆基因,参与绿盲蝽耐受高温、杀虫剂等胁迫反应。为了明晰绿盲蝽取食不同寄主植物后其体内抗逆基因的表达特征,本试验通过荧光定量PCR和Western杂交技术,重点分析AlHSP90基因及其蛋白在绿盲蝽取食不同寄主植物后的表达谱。结果表明:绿盲蝽雌、雄成虫取食Bt棉、常规棉后,AlHSP90表达量显著高于取食其他寄主植物(P<0.01);与对照四季豆相比,取食茼蒿之后AlHSP90表达量无显著变化(P>0.05)。雌成虫取食Bt棉、常规棉后,AlHSP90表达量分别比对照上升了1.77、1.74倍;而雄成虫取食Bt棉、常规棉后,AlHSP90表达量则分别比对照上升了1.98、1.94倍。取食不同寄主植物之后,AlHSP90蛋白的表达谱结果与其基因的表达谱结果高度相似。绿盲蝽雌、雄成虫取食Bt棉、常规棉后,AlHSP90蛋白表达量显著高于取食其他寄主植物(P<0.01);而取食茼蒿之后,AlHSP90蛋白表达量无显著变化(P>0.05)。由此可见,AlHSP90基因是绿盲蝽适应棉花取食过程中重要的抗逆基因,并在绿盲蝽寄主转换过程中起到重要作用。 The heat shock protein 90 gene A1HSP90 is an important stress-resistant gene in Apolygus lucorum, involved in the re- sistance or tolerance ofApolygus lucorum to heat shock or pesticide stresses. To determine the expression profile of the stress-re- sistant gene in Apolygus lucorum after adults were fed on different host plants, this study set A1HSP90 as the objective gene and analyzed the expression profiles of the A1HSP90 gene and protein in Apolygus lucorum using real-time PCR and westem blot. The expression of A1HSP90 increased significantly when female and male adults ofApolygus lucomm were fed on Bt and convention- al cottons, and was significantly higher than with other host plants (P 〉0.01). However, no significant difference of A1HSP90 ex- pression was found between garland chrysanthemums and the control (P 〉 0.05). After female adults ofApolygus lucorum were fed on Bt cottons and conventional cottons, the expression of A1HSP90 was 1.77 and 1.74 times higher than the control, respective- ly. After the male adults ofApolygus lucorum were fed on Bt cottons and conventional cottons, the expression of AIHStO0 was 1.98 and 1.94 times higher than the control, respectively. The expression file results of the A1HSP90 protein were similar to those of the AIHSIO0 gene, which confirmed that after female and male adults of Apolygus lucorum were fed on Bt and conventional cottons, the expressions of the A1HSP90 protein were significantly higher than with other host plants (P 〉0.01). Furthermore, no significant difference in the A1HSP90 protein expression was found between garland chrysanthemums and the control (P 〉 0.05). Therefore, it was determined that AIHSP90 is an important stress-resistant gene for Apolygus lucorum adults fed on cottons, and that it plays an important role in the host shift process ofA. lucorum.
