恶性腹腔积液来源的树突状细胞与CIK共培养后对结肠癌细胞体外杀伤作用的研究

摘要 目的探讨恶性腹腔积液来源的树突状细胞(DC)与CIK细胞共培养后对SW620结肠腺癌细胞的体外杀伤作用。方法分离15例结肠癌患者的腹腔积液培养获得成熟DC,分离健康人外周血培养获得CIK细胞,DC细胞与CIK细胞共培养,流式细胞仪鉴定表型,MTT法检测各组对结肠癌细胞的体外杀伤作用。结果恶性腹腔积液中存在DC前体细胞,经体外细胞因子诱导培养后可获得成熟的DC细胞,高表达HLA-DR、CD80、CD83、CD86分子。DC与CIK共培养对SW620结肠腺癌细胞的杀伤作用较单独CIK组有明显差异(P<0.05)。结论腹腔积液中存在DC前体细胞在体外诱导培养可获得功能正常的成熟DC细胞,与CIK共培养后可增强其体外杀伤毒性。 Objective To explore the anti-tumor effect of dendritic cells (DC) deriving from malignant ascites co- cultured with CIK against SW620 colon adenoearcinoma cell in vitro. Methods Mature DCs were obtained by density gradi- ent eentrifugation from ascites of 10 patients with colon cancer. CIK cells were prepared routinely from human peripheral blood monomuclear ceils (PBMC). CIK ceils were co-cultured with DC. Flow cytometer was used to explore the immunophenotypes, and the anti-tumor effect was examined by MrlT assa in vitro. Results There were DC precursor cells in malignant aseites, DCs of ascites had the typical morphology after extracorporeal cultivation, which showed the high expressions of HLA-DR, CD83, CDSO, CD86. DCs co-cultured with CIK showed significant inhibitory effects on the colon cells compared with CIK alone (P 〈 0.05 ). Conclusion DC precursor cells deriving from malignant ascites were cultured in vitro can be obtained mature DC cells ,which can enhance the anti-tumor effect of CIK cell when they are co-cultured with CIK.
