基于含药肠吸收液法的复方丹参片对大鼠离体胸主动脉环的作用及机制初步研究

Experimental Study on Influence in Vascular Rings Tension of Compound Danshen Tablet at Rat Thoracic Aorta in vivo Based on the Method of TCM Prescription-Containing Intestinal Absorption

目的初步探讨含药肠吸收液方法用于中药复方体外药理作用与作用机制研究的可行性。方法翻转囊法制备复方丹参片含药肠吸收液,采用大鼠离体胸主动脉环灌流模型,观察累积浓度含药肠吸收液对基础状态、KCl预收缩、有钙/无钙液苯肾上腺素(PE)预收缩血管环的作用;对PE和KCl引起的依赖于细胞内钙和细胞外钙收缩反应的影响;吲哚美辛、L-NAME对其舒张血管作用的影响,并与硝苯地平比较。结果复方丹参片含药肠吸收液对基础状态血管环无明显影响;可剂量依赖性抑制KCl、有钙/无钙液PE预收缩的血管;可拮抗PE和KCl诱发的内钙/外钙依赖性收缩反应;而L-NAME、吲哚美辛不能阻断其舒张血管作用。结论复方丹参片含药肠吸收液可主要通过阻断钙通道产生舒张血管平滑肌的作用,提示含药肠吸收液方法可用于中药复方体外药理研究。

Objective To investigate the feasibility of TCM prescription - containing intestinal absorption method used on pharma- cological effects and mechanism study preliminary in vitro. Methods Intestinal absorption of Compound Danshen Tablet （CDST） was prepared by the reverted gut sac experiments in vitro, the study was performed with the model of isolate rat thoracic aorta rings in organ bath. The effect of accumulated CDST - containing intestinal absorption on aorta rings in resting tension, pre - constric- ted with KCl, pre - constricted with phenylephrine （PE） under the Ca2 ＋ - contained and Ca2 ＋ - free bath solutions were ob- served and were compared with nifedipine. The effect on the contraction of vascular smooth muscle depending on the intracellular calcium and extracellular calcium induced by PE or KC1, and the influence in its vascular contraction induced by NA in the pres- ence of L - NAME or indomethacin were also observed. Results Accumulated CDST - containing intestinal absorption had no sig- nificant effects on aortic rings in resting tension, but had concerned ration - dependent relaxation of vascular rings pre - constric- ted with KCl, or pre - constricted with PE under the Ca2 ~ - contained and Ca2 ＋ - free. It also had significant effects on the aorta contraction induced by intracellular calcium and extracellular calcium, the relaxant effect could not be attenuated by L - NAME or indomethaein. Conclusion CDST -containing intestinal absorption had relaxant effect on vascular smooth muscle, which is mainly connected with calcium channel blocking. The results indicate that the method of TCM prescription - containing intestinal absorption can be used on pharmacology study in vitro.
