摘要
目的研究神经干细胞具有自我更新、未分化和多向分化潜能,冬虫夏草中的氨基酸、核苷和糖蛋白等能诱导神经干细胞的定向分化。方法取新生24 h内的Wistar大鼠大脑海马区分离培养神经干细胞三代后,加入低、中、高剂量冬虫夏草含药血清与对照血清进行培养,观察其对神经干细胞影响,通过MTT检测神经细胞生长曲线,免疫组化法和RTPCR半定量检测神经干细胞向神经元分化的情况。结果新生大鼠海马分离培养的神经克隆球,经免疫细胞化学染色检测为Nestin阳性细胞。NSC贴壁分化的细胞呈NSE、GFAP阳性。冬虫夏草低、中药物实验组诱导神经干细胞分化为神经元的阳性细胞明显高于对照组,且呈量效依赖关系,高浓度药物实验组与对照组没有明显差别。结论冬虫夏草药物血清可体外诱导神经干细胞向神经元方向分化,在一定范围内存在量效依赖关系。
Objective To study the self - renewal of neural stem cells, undifferentiated and multi - differentiation potential of Cordyceps sinensis in amino acids, nucleotides and sugar protein can induce direct differentiation of neural stem ceils. Method- s Isolate newborn within 24 hours of the wistar rat hippocampus and culture neural stem cells in three generations. Then add low, medium and high doses of Cordyceps sinensis containing serum and control serum culture medium to observe its neural stem cells affected by mtt. Detect nerve cell growth Curve, immunohistochemistry and semi - quantitative RT - PCR detection of neural stem cells and then differentiate them into neurons in the situation. Results The isolated hippocampal nerve clone ball, detected by im- munocytochemistry as Nestin - positive ceils. The differentiated cells adhering to NSC were NSE, GFAP positive. Cordyceps sinensis is low, the positive cells differentiated by experimental group of drug induced neural stem cells was significantly higher, and the dose was in proportion with its effect; There is no significant difference between experimental group and control group for the high concentrations of drugs. Conclusion The serum Cordyceps can induce neural stem ceils to differentiate into neurons, and to some extent, the effects depend on the dose.
