摘要
以根癌农杆菌感染的锦橙Kanr 抗性芽为试材 ,利用改良MS液体培养基湿润滤纸桥培养进行生根研究 ,结果表明 :2 5 0mg/L的IBA处理抗性芽基部 2 0min后 ,培养在含蔗糖浓度为 30g/L的改良MS液体培养基湿润滤纸桥上 ,30天后 ,生根率可以达到 78% ,平均根数 1.7条 ,平均根长 3.5 3cm 。
An experiemnt was made with karamycin-resistant shoots of Jin Cheng orange ( Citrus sinensis Osbeck) obtained from Agrobacterium- mediated infection to investigate the optimum conditions for their rooting. The base of the shoots was treated with 250 mg/L IBA for 20 min. And then placed onto the filter paper-bridge that was soddened with a modified MS liquid medium supplemented with 30 g/L sucrose. After 30 days' culture, the rooting rate of the shoots was up to 78%, with an average root number per shoot and root length of 1.7 and 3.53 cm, respectively. Such a procedure is, therefore, recommended for improving the regeneration rate of transgenic citrus.
