摘要
目的建立HPLC法测定病毒合剂中黄芩苷、绿原酸和连翘酯苷A的方法。方法 Shim-pack VP-ODS色谱柱(150mm×4.6 mm,5μm),流动相为0.5%磷酸水溶液–乙腈,采用梯度洗脱,检测波长为300 nm,体积流量为1.0 mL/min,进样量为10μL。结果黄芩苷、绿原酸和连翘酯苷A分别在9.84-82μg/mL(r=0.999 8)、2.64-22.0μg/mL(r=0.999 9)和4.32-36.0μg/mL(r=0.999 9)线性关系良好,平均回收率分别为99.94%、101.1%、99.77%,RSD值分别为1.44%、1.68%、1.44%(n=6)。结论本法操作简便、结果准确、重复性好,可用于病毒合剂的质量控制。
Objective To establish an HPLC method for the determination of baicalin, chlorogenic acid, and forsythoside A in Bingdu Mixture. Methods HPLC was carried out on a Shim pack VP-ODS column(150 mm × 4.6 mm, 5 μm) with 0.5% phosphoric acid- acetonitrile as mobile phase for gradient elution. The detection wavelength was set at 300 nm. The injection volume was 10 μL at the flow rate of 1.0 mL/min. Results The liner ranges of baicalin, chlorogenic acid, and forsythoside A were 9.84 — 82 μg/mL(r = 0.999 8), 2.64 — 22.0 μg/mL(r = 0.999 9) and 4.32 — 36.0 μg/mL(r = 0.999 9); The average recoveries were 99.94%, 101.1%, and 99.77%, with RSD values of 1.44%, 1.68%, and 1.44%(n = 6), respectively. Conclusion The established method is simple, accurate, and reproducible and could be used for the quality control of Bingdu Mixture.
