摘要
为优化浒苔膳食纤维提取工艺,采用单因素和响应面试验研究酶解温度、酶解时间、pH和复合酶质量比对浒苔膳食纤维提取得率的影响。利用响应面法(RSM)建立二次回归模型,对4因素进行优化组合,得到浒苔膳食纤维最佳工艺条件为酶解温度55℃、酶解时间91 min、pH 7.7、复合酶质量比20∶1(蛋白酶与纤维素酶质量比为20∶1),此时,膳食纤维实际提取得率为91.45%。膳食纤维功能特性初步评价结果表明,膳食纤维具有较好的持水力和膨胀力,分别为11.2 mL/g和8.52 mL/g;同时具有较强的吸附葡萄糖及胆固醇的能力,其最大吸附值分别为20.02 mg/g和22.52mg/g。方法提取的浒苔膳食纤维具有良好的品质特性,可作为降血糖、降血脂功能食品的潜在来源。
To obtain the optimum enzymolysis extraction technology of dietary fiber fi'om Enteromorpha, the single-factor tests and Response surface methodology (RSM) were employed to optimize the extraction conditions, including protease/ cellulose ratios, reaction time, reaction temperature and pH. Quadratic regression model was established by RSM, combination of four factors was optimized. The optimum extraction conditions were: temperature 55 ~C, time 91 min, pH 7.7 and composite enzyme ratio 20 : 1. Under optimum conditions, the extraction yield of dietary fiber was 91.45% and they had good water holding capacity and expansive capacity, and the values were 11.2 mL/g and 8.52 mL/g. The adsorption results showed that the adsorption capacities of glucose and cholesterol were strong. The maximum adsorption values were 20.02 mg/g and 22.52 mg/g, respectively. With the good properties, the Enteromorpha dietary fiber was suitable for hypoglycemic functional food and cholesterol-lowering functional food.
