超声联合超微粉碎法提取金针菇多糖新探究 被引量:15

A New Research on Ultrafine Comminution Combining Ultrasonic Extraction of Flammulina velutipes Polysaccharides

摘要 为提高金针菇多糖的提取率,采用超微粉碎技术处理金针菇原料,在单因素试验的基础上,选取超微粉碎粒度、超声功率、超声时间进行Box-Behnken中心组合设计,以金针菇多糖提取率为响应值。通过响应面优化,得到最优工艺为:超微粉碎粒度395.71目、超声功率380.03 W、超声时间24.56 min,该试验条件下金针菇多糖提取率为20.76%,经修正后在超微粉碎粒度400目、超声功率380 W、超声时间25 min条件下进行验证,实际提取率为20.52%,实际值接近理论预测值,表明超声联合超微粉碎可以显著提高金针菇多糖的提取率。 In order to improve the extraction ratio of Flammulina velutipes Polysaccharides (FVP), ultmfine comminution was firstly applied into the material pretreatment. Based on single factor and extraction ratio as response values, granularity, ultrasonic power and ultrasonic time as response factors, study was conducted by Box-Behnken central composite experimental design. The optimum conditions of extraction obtained by response surface methodology were as followed: granularity at 395.71 mesh, ultrasonic power of 380.03 W, and ultrasonic time of 24.56 min. Under these conditions, the extraction ratio was 20.76%. The extraction ratio 20.52% was measured with granularity at 400 mesh, ultrasonic power of 380 W, ultrasonic time of 25 min after modification, which was close to the predict value. The model indicated that ultrasonic and ultrafine comminution has a significant impact on extraction ratio of FVP.
