Effect of WY14643 on vascular endothelial function in hypertensive rats

摘要 目的观察WY14643对高血压大鼠血管内皮的作用机制。方法通过将38-42周的自发性高血压大鼠SHR分离获取主动脉,水浴在10 ml的组织浴槽中。随机分为2组,SHR组9只使用蒸馏水处理作为对照,WY14643组10只使用WY14643进行干预,搜集浴槽中的液体,检测血管舒张因子的变化,检测血管组织中内皮型一氧化氮合酶(endothelial nitric oxide synthase,eNOS)的改变。结果经过WY14643处理后,与SHR组相比,WY14643组NO(23.4±3.2)μmol/L和PGI2(前列环素)(413.5±113.6)ng/L的含量明显升高,差异具有统计学意义(P<0.05)。结论 WY14643对高血压大鼠血管内皮具有保护作用。 Objective To observe the effect of WY14643 on vascular endothelium in rats with hypertension. Methods We selected spontaneous hypertensive rats(SHR) of 38-42 weeks, isolated the aorta and bathed it in 10 ml tissue bath. We randomly divided them into 2 groups: rats of SHR group was just treated with distilled water as control group, 10 rats of WY14643 group was only treated with WY14643 as intervention group.The liquid in bath was collected to detect the changes of vasodilator and eNOS in vascular tissue. Results NO and PGI2(prostacyclin) levels and eNOS(nitric oxide synthase) expression were significantly increased after WY14643 treatment.There was a statistically significant difference (P〈0.05) between the two groups. Conclusions WY14643 plays a protective role on vascular endothelium in rats with hypertension.
