摘要
用夏芳♂、夏sch♂、秋白♂、秋sch♂两对近等位基因系为材料,采用随机引物进行 PCR扩增。通过90个随机引物,在夏sch♂中得到OPD02-400bp多态性特异带,用秋白、秋sch进 一步验证确认:OPD02-400bP是sch基因的 RAPD标记。将获得的分子标记 DNA片段回收纯化 后,克隆到质粒pBluescript-IISK载体上。
PCR amplification was made with arbitrary primers, using two pairs of near - isogenic lines(Xiafang ♂ and Xia sch ♂, and Qiubai ♂ and Qiu sch ♂ ) of silkworm as the experiment materials. A polymor- phic specific band, OPD02 - 400bp, was obtained with 90 arbitrary primers in Xia Sch ♂. A further ex- periment with Qiubai and Qiu sch confirmed that OPD02 - 400bp is the PAPD marker for the sex - linked chocolate gene sch. The DNA fragments of the molecular marker were recovered, purified and cloned onto the carrier - plasmid pBluescript - II SK.
