小鼠感染细粒棘球蚴早期TGF-β1表达分析 被引量:3

The expression of TGF-β1 increases in the early stages of infection with Echinococcus granulosus in mice

摘要 目的观察细粒棘球蚴早期感染BALB/c小鼠TGF-β1的表达情况。方法用细粒棘球蚴感染BALB/c 小鼠,分别于感染后第1、3、5、7、9、12d处死,取其脾细胞和外周血,采用酶联免疫法检测外周血细胞因子TGF-β1的含量,采用qRT-PCR法检测TGF-β1基因的表达量。结果实验鼠Eg感染后第1~12d,外周血TGF-β1的含量为2695.79~3055.09ng/ml,与对照组比较差异有统计学意义(P〈0.05);TGF-β1 mRNA相对表达量为0.029~0.060,与对照组比较差异有统计学意义(P〈0.05)。TGF-β1及其mRNA表达量均有随着感染时间延长譬增高的趋势。结论TGF-β1在小鼠细粒棘球蚴感染早期表达增加,可能有利于细粒棘球蚴的免疫逃逸。 Objective To observe the changes in TGF-β1 in the early stages of Echinococcus granulosus infection in mice. Methods BALB/c mice were infected with E. granulosus, and spleen cells and peripheral blood were collected 1, 3, 5, 7, 9, and 12 days post-infection. The cytokine TGF-β1 levels in peripheral blood were analyzed using ELISA; the relative expression of TGF-β1 mRNA was detected using quantitative RT PCR. Results From 1 to 12 days post-infec- tion, the levels of TGF-β1 in peripheral blood were significantly higher than those in the control group (P〈0.05). The levels of TGF-β1 ranged from 2 695.79 to 3 055. 09 ng/mL The relative expression of TGF-β1 mRNA was 0. 029 to 0. 060, which was statistically greater (P〈0.05) than that in the control group. Conclusion The levels of TGF -β1 in peripheral blood were found to increase in mice during early infection with E. granulosus TGF-β1 facilitates evasion of the host's immune system by E. granulosus.
