摘要
目的:观察止哮汤对哮喘大鼠气道重塑及肺组织增殖细胞核抗原(PCNA)的影响。方法:选用健康Wistar大鼠60只,雌雄不拘,体重(200±20)g,随机分为6组:正常对照组(A组),哮喘模型组(B组),地塞米松对照组(C组),中药低剂量组(D组)、中剂量组(E组)、高剂量组(F组)共6组,10只/组。利用卵蛋白加氢氧化铝复制哮喘大鼠模型,各组大鼠均经末次激发24 h后处死。取左肺在矢状面取材按常规方法作病理切片及HE染色。光镜下观察哮喘大鼠的气道改变;免疫组化法分别测定大鼠肺组织PCNA蛋白的表达。结果:模型组大鼠气道壁及气道平滑肌面积明显增厚,气道壁面积、气道平滑肌面积以及支气管平滑肌细胞数经支气管内周长标准化后,亦明显高于对照组,表明在哮喘急性加重期哮喘大鼠已出现了气道平滑肌的增殖和气道重建,证明了在哮喘急性期存在着气道重塑。正常对照组PCNA呈阴性表达,模型组PCNA表达呈强阳性,强于正常对照组;地塞米松组、中药各剂量组PCNA表达均弱于模型组;中药低剂量组强于地塞米松组;中药中、高剂量组则与地塞米松组无差异;中药中、高剂量组均弱于中药低剂量组;中药高剂量组与中药中剂量组表达无差异。结论:止哮汤可下调哮喘大鼠肺组织中PCNA的表达,减轻气道重塑,高、中剂量组疗效较好,与地塞米松效果相当。
Objective : To observe the influence of Antiasthma Decoction on acute asthmatic rat dexamethasone and PC- NA in lung tissue. Method : 60 healthy Wistar rats of either sex, weighing (200 ± 20) g, were randomly divided into six groups : the normal control group ( A), asthma model group ( B), dexamethasone control group ( C), low dose of Antiasth- ma Decoction group ( D), medium dose of Antiasthma Decoction group ( E), high dose of Antiasthma Decoction group ( F), six groups in total, 10 for each group. We used aluminum hydroxide to copy ovalbumin rat model of asthma. The rats were sacrificed after the last challenge 24 h. The left lung in the sagittal plane drawn by conventional methods were slicedfor HE staining. Light microscope showed airway changes in rats with asthma, and the acute asthma model was successfully verified. PCNA protein in lung tissue was measured by Immunohistochemical method. Result : Model rats airway wall area and airway smooth muscle were thickening. The airway wall area, airway smooth muscle area and the number of bronchial smooth muscle cells became higher within the standardized perimeter. It showed an acute exacerbation of asthmatic rats which emerged airway smooth muscle proliferation and airway reconstruction, indicating acute airway remodeling existed. Normal group~ PCNA was negative and model group was positive and stronger than that of normal group. Dexamethasone group and Chinese medicine dose groups'PCNA expressions were weaker than that of the model group. PCNA expression in low dose group was stronger than that in the dexamethasone group. Chinese medicine high dose group and dexametha- sone group had no difference. Chinese medicine and high dose group's PCNA expressions were lower than that of low dose group. Chinese medicine high and medium doses groups showed no difference. Conclusion:The Antiasthma Decoction can reduce PCNA expression in asthmatic lung tissue and airway remodeling. The high and medium dose groups have the effect as good as dexamethasone.
