摘要
研究了香菇菌和酵母菌共生发酵葡萄渣和麸皮生产纤维素酶的条件。香菇菌和葡萄酒酵母菌可以在PDA液体培养基中共生。并产生纤维素酶。通过正交试验法确定了香菇菌与葡萄酒酵母菌共生的主要基质最佳组合:葡萄渣、蔗糖和麸皮质量分数均为3%,硫酸镁和磷酸二氢钾质量分数均为0.15%。正交试验法确定的2种菌共生发酵产生纤维素酶的最佳条件为:发酵产酶温度27℃,pH5.5,共生发酵3~5d,纤维素酶活力为3.2U/g,香菇菌生物量为21.4g/L,酵母菌细胞计数为3.61×10^6个/mL。比较试验证明共生发酵产生的纤维素酶与香菇菌单菌发酵结果一致。
Culture conditions of Lentinula edodes together with yeast to produce cellulase in grape residue-bran liquid medium were studied. Dentinula edodes and yeast could coexist in PDA medium to produce cellulase. The optimum formula were confirmed as follows: grape pomace 3%, wheat bran 3%, MgSO4 0.15%, KH2PO4 0.15%. The optimum condition of Lentinula edodes together with yeast to produce cellulase were also confirmed: 27 ℃, pH 5.5, fermentation time 3-5 d, cellulose enzyme activity 3.2 U/g. Under this condition, the Lentinula edodes biomass could reach 21.4 g/L and the amount of yeast cells was 3.61 × 10^8 cells/mL.
