摘要
目的:通过检测半夏泻心汤对幽门螺杆菌主要毒力因子CAGA、VacA和γ-GGT的mRNA表达量的影响,探求半夏泻心汤抗幽门螺杆菌的作用机制。方法:采用琼脂稀释法进行抑菌实验,观察半夏泻心汤对幽门螺杆菌的抑菌效果;运用分子生物学技术RT-PCR检测半夏泻心汤对幽门螺杆菌的主要毒力因子CAGA、VacA和γ-GGT的mRNA表达量的影响。结果:半夏泻心汤对幽门螺杆菌的最低抑菌浓度即MIC值为25mg/mL;受试幽门螺杆菌的主要毒力因子CAGA、VacA和γ-GGT的mRNA表达量在半夏泻心汤的影响下均明显下调。结论:半夏泻心汤抗幽门螺杆菌相关性胃肠道疾病的作用机制与半夏泻心汤对幽门螺杆菌的直接抑制作用以及对其主要毒力因子CAGA、VacA、γ-GGT的毒力作用的削弱密切相关。
Objective:To examine the effect of Banxia Xiexin decoction on the mRNA expression of main virulence factors of Helico- baeter pylori (HP) ( CagA,VaeA, and y - GGT) and to investigate the anti - HP mechanism of Banxia Xiexin decoction. Methods : An- tihaeterial test was performed by agar dilution to observe the inhibitory effect of Banxia Xiexin decoction on HP. The effect of Banxia Xiexin decoction on the mRNA expression of main virulence factors of HP (CagA,VaeA,and y- GGT) was evaluated by RT- PCR. Results :The minimal inhibitory concentration of Banxia Xiexin decoction for HP was 25 mg/ml. The mRNA expression of CagA, VacA, and y - GGT was significantly down - regulated in the presence of Banxia Xiexin deeoetion. Conclusion : The action meehanism of Banx- ia Xiexin deeoetion in the treatment of HP - related gastrointestinal diseases is closely related to its direct inhibitory efteet on HP and capability of attenuating the virulence of CagA, VacA, and y - GGT.
