滋肾平颤颗粒对异动症大鼠行为学及黑质纹状体DAT和VMAT2基因表达的影响 被引量:7

摘要 目的:观察滋肾平颤颗粒对异动症(levodopa-induced dyskinesias,LID)大鼠行为学和黑质纹状体多巴胺转运体(dopamine transporter,DAT)及囊泡单胺转运体2(vesicular monoamine transporter2,VMAT2)基因表达的影响。方法:采用6-羟基多巴胺(6-hydroxydopamine,6-OHDA)注射于大鼠脑右侧黑质造成偏侧帕金森病(Parkinson’s disease,PD)模型,进一步对PD大鼠予以左旋多巴/苄丝肼制作异动症模型。实验设立正常对照组、模型组、中药干预组、中止给药对照组和中止给药+中药干预组,分别处理或给药1个月后观察药物对LID大鼠异常不自主运动(abnormalinvoluntary movement,AIM)评分的影响,运用实时定量PCR检测黑质纹状体DAT及VMAT2 mRNA表达。结果:与模型组和中止给药对照组比较,中药干预可明显减少异动症大鼠的AIM积分;荧光实时定量PCR结果显示,模型组大鼠DAT mRNA以及VMAT2 mRNA表达率明显低于正常对照组及其他各组,中药干预组与模型组,中止给药+中药组与中止给药组比较,两种mRNA表达均上调,其中VMAT2增高的趋势更明显。结论:中药可以有效缓解异动症症状,上调DAT mRNA以及VMAT2 mRNA表达。
