摘要
探明马尾松种子的萌发机理,为贵州马尾松种子育苗提供参考,采用酶联免疫间接法对马尾松种子萌发期胚乳内的GA3、IAA、ABA、ZR、SOD、POD和淀粉酶含量进行测定,研究马尾松种子萌发过程中各种内源激素和酶活性的变化。结果表明:1)在马尾松种子萌发期,胚乳内的GA3、IAA、SOD、POD和淀粉酶含量随种子萌发时间的延长逐渐升高,0h到80h时的增幅分别为61.7%、93.6%、48.3%、49.5%和26.7%;ZR含量呈先升高后降低变化趋势,并在60h时达最高,为30.29ng/g;ABA含量呈逐渐降低趋势,由0h时的48.13ng/g降到80h时的24.24ng/g。2)马尾松种子胚乳中的GA3/ABA、IAA/ABA和ZR/ABA随种子萌发培养时间的延长呈逐渐升高趋势,并在培养结束时达到最大值;GA3、IAA、SOD、POD、淀粉酶含量的升高有助于马尾松种子萌发,ABA含量的升高则抑制马尾松种子萌发。因此,协调好马尾松种子各内源激素的变化对其种子顺利萌发有重要作用。
To discuss the mechanism of P .massoniana seed during germination stage and provide references for cultivating P .massoniana seedlings,the changes of GA3 ,IAA,ABA,ZR contents and SOD,POD and amylase were studied by indirect ELISA method.Results:1)GA3 ,IAA,SOD,POD and amylase presented an gradually increasing trend in initial phase,with the growing rates of 61.7%, 93.6%,48.3%,49.5% and 26.7% respectively.ZR first increased and then decreased,and reach the maximum of 30.29 ng/g at 60 h.ABA gradually decreased from 48.13 ng/g to 24.24 ng/g.2)Values of GA3/ABA,IAA/ABA and ZR/ABA gradually increased with culture time prolonging and reached the maximum in the end of cultivation.The enhancement of GA3 ,IAA,SOD,POD and amylase were beneficial for P .massoniana seed germination.While the enhancement of ABA inhibited the germination. So the coordination of various endogenous hormones played an important role in seed germination.
