水晶布兰卡百合鳞片的组织培养及快速繁殖条件 被引量:2

摘要 为筛选水晶布兰卡小鳞茎诱导、增殖、生根的最佳培养基配方和最佳培养条件,以水晶布兰卡百合鳞片为外植体,研究了从外植体诱导分化小鳞茎的培养基配方、暗处理时间、小鳞茎增殖的最佳培养基配方和最佳切割方式,生根的最佳激素配比。结果表明:在初代培养中,不同培养基的小鳞茎诱导系数都较高,最佳诱导培养基为MS+6-BA 1.5mg/L+NAA 0.5mg/L,诱导系数为3.63;暗培养有利于小鳞茎诱导,且暗培养40d小鳞茎诱导系数比暗培养30d的高;在增殖培养中,带基部鳞片切割方式培养于4种不同的培养基均可获得较高的增殖系数,增殖系数最高的培养基配方为MS+6-BA 0.5mg/L+NAA 0.25mg/L+KT 0.25mg/L,达3.67;在生根培养中,诱导小鳞茎生根最佳培养基为1/2MS+NAA 0.4mg/L+IBA0.1mg/L,生根系数达5.2。 In order to screen the best culture medium and the best culture conditions for the differentiation,multipulication and rooting of small bulb of L.casa.In this paper the scales of L.casa was used as explants,the small bulb from initially generation was cut by different way,different culture medium was used for the initially generation culture,the multipulication culture and the root inducing. The results showed that in the initially generation culture the small bulb differentiation rate of different culture medium was quite high,the best medium for small bulb differentiation was MS+6-BA 1.5 mg/L+NAA0.5 mg/L with differentiation coefficient of 3.63.Dark training was advantageous to the small bulb induction,and dark training 40 d was better than that of dark training 30 d.In multipulication culture,the multipulication rate of the small bulb with base flake cutting way cultured on four kinds of different culture medium was quite high, the best medium for multipulication of the small bulb was MS + 6-BA 0.5 mg/L +NAA0.25 mg/L + KT0.25 mg/L with multipulication coefficient of 3.67.In rooting culture, the best medium for inducting small bulb root was 1/2 MS + NAA0.4 mg/L + IBA0.1 mg/L,the coefficient of rooting rate was 5.2.
