五没食子酰基葡萄糖对瘢痕疙瘩成纤维细胞增殖与凋亡的影响 被引量:7

摘要 目的:确定五没食子酰基葡萄糖对瘢痕疙瘩细胞增殖与凋亡的影响。方法:用细胞生长实验,细胞周期分析,细胞姬姆萨染色比较五没食子酰基葡萄糖、五倍子单宁酸和秋水仙碱对瘢痕组织成纤维细胞,Hacat细胞和肿瘤细胞在增殖与凋亡方面的影响。结果:五没食子酰基葡萄糖对瘢痕疙瘩成纤维细胞和黑素瘤A375细胞的增殖有较强的抑制作用,对成纤维细胞的作用呈时间和剂量依赖性,对HaCat细胞的增殖呈双相作用,低浓度为促进作用,高浓度为抑制作用,可使增殖受抑制细胞的细胞周期发生改变,表现为S期阻滞。凋亡细胞显著增加。结论:五没食子酰基葡萄糖对瘢痕疙瘩成纤维细胞有抑制增殖和促进凋亡的作用,对治疗瘢痕疙瘩可能是一个具有开发潜力的活性物质。 Objective: To determine the effect of pentagalloyl glucose on the proliferation and apoptosis of keloid fibroblasts. Methods: The impact of pentagalloyl glucose, gallotannin and colchicine on the prolifera- tion and apoptosis of keloid fibroblasts, HaCat cells and melanoma A375 cells was determined by cell growth test, cell cycle analysis and Giemsa stain method. Results: Pentagalloyl glucose had a relatively strong inhibi- tory effect on keloid fibroblasts and melanoma A375's proliferation. It significantly inhibited fibroblast prolifer- ation in a dose- and time-dependent manner. It had dual roles on the proliferation of HaCat. It enhanced the proliferation in low concentration and an inhibitory effect in high concentration. It inhibited the growth of der- mal fibroblasts by interfering with cell cycle progression from S phase to G2/M phase. Cell Giemsa stain showed that there were increase in cells apoptosis. Conclusion: Pentagaltoyl glucose can inhibit proliferation and enhance apoptosis of keloid fibroblasts. It can be a potential active substance in the treatment of keloid.
