摘要
目的观察大鼠孕期酒精暴露对子代胆碱能神经系统的影响。方法妊娠SD母鼠酒精2g·kg-1·d-1(模型组)或等体积蒸馏水(对照组)灌胃至分娩前。8周龄子代鼠进行水迷宫实验测定学习记忆行为,乙酰胆碱转移酶免疫组化和乙酰胆碱酯酶组织化学检测基底前脑胆碱能神经元和海马胆碱能纤维的变化。结果与对照组相比,模型组学习记忆能力下降(P<0.05),基底前脑胆碱能神经元数目和海马乙酰胆碱能纤维终末减少(P<0.01)。结论孕期酒精暴露可导致子鼠隔-海马胆碱能神经系统损害。
Objective To investigate the effect of prenatal ethanol exposure on the cholinergic system of offspring rats. Methods Pregnant SD rats were given gastric gavage with ethanol 2 g · kg-1· d-1 (group A) or same volume of distilled water (group B) throughout gestation. The 8-week-old offspring rats were subjected to behavioral tests. Choline acetyltransferase inmlunobistochemistry and acetylcholinesterase histochemistry were performed to detect cholinergic neurons in the basal forebrain and cholinergic terminals in the hippocampus, respectively. Results Compared with group B, the abilities of learning and memory of the offspring rats were impaired (P〈0. 05), and the cholinergic soma in the basal forebrain and cholinergic terminals in the hippocampus were decreased in group A (P〈0. 01 ). Conclusion Prenatal ethanol exposure may impair the septohippocampal cholinergic system in offspring rats.
