摘要
目的研究苯并咪唑对体外培养大鼠睾丸支持细胞波形蛋白和微管蛋白表达的影响。方法取18—20日龄Wistar雄性大鼠睾丸支持细胞,通过Tris-Hcl低渗处理进行细胞纯化,用MTT法检测阴性对照组、溶剂对照组、苯并咪唑剂量组(0.1、1、10和100μg/m1)中大鼠睾丸支持细胞的生长抑制率。用免疫细胞化学法检测细胞中波形蛋白、α/β-微管蛋白的表达。结果10和100μg/ml苯并咪唑剂量组在24、48和72h时均明显抑制了支持细胞的增殖;10和100pg/ml苯并咪唑剂量组中支持细胞波形蛋白、α/β-微管蛋白表达明显减少。结论苯并咪唑对体外培养大鼠支持细胞毒作用明显,抑制睾丸支持细胞的增殖,影响波形蛋白、α/β-微管蛋白的表达。
Objective To study the effects of benzimidazole on the expression of vimentin and microtubule proteins in rat testicular Sertoli cells in vitro culture. Methods Sertoli cells were harvested from rat testes aged 18 - 20 days, and then purified by Tris-Hcl low permeability treatment. The growth rates of Sertoli cells were determined with MTT assay in the negative control group, the solvent control group and benzimidazole dose groups. The expression of vimentin and microtubule proteins was detected by immunocytochemistry method. Results The proliferation of Sertoli cells was significantly inhibited in 10 and 100 μg/ml benzimidazole groups in 24, 48 and 72 h. The expression of vimentin and microtubule proteins in Sertoli cells in 10 and 100 μg/ml benzimidazole groups was significantly decreased. Conclusion Benzimidazole has obvious toxic action to rat Sertoli ceils in vitro culture, inhibites the proliferation of Sertoli cells, affects the expression of vimentin and α/β-tubulin in Sertoli cells.
