摘要 目的:研究MRL小鼠IL-10R1对B淋巴细胞功能的影响。方法:分别将克隆有MRL小鼠和C57BL/6小鼠IL-10R1基因的载体转染至Ba/F3细胞稳定表达,经IL-10刺激后,用MTT法检测细胞增殖情况,用流式细胞术检测CD16/32和LECAM-1的表达情况。结果:随着IL-10的浓度增加,表达MRL IL-10R1的B细胞的增殖能力递增,而表达C57BL/6 IL-10R1的B细胞递减;接受IL-10刺激后表达MRL IL-10R1的B细胞CD16/32表达水平较表达C57BL/6 IL-10R1的B细胞低,LECAM-1表达水平前者却较后者高。结论:MRL小鼠IL-10R1丧失了对B细胞活化、增殖及趋向性迁移的抑制作用,这可能与系统性红斑狼疮的发生及发展有关。 Objective:To investigate that the IL-10R1 of MRL mouse affects the functions of B lymphocytes. Methods: The vectors with the iL-10R1 gene of MRL or C57BL/6 mouse were transferred into Ba/F3 cells. After transfection, cell proliferation curve was evaluated by MTF. The expressions of CD16/32 and LECAM-1 were performed by FCM. Results: With the increase of IL-10 con- centration the proliferation ability of B cells with MRL IL-IOR1 was increased gradually, but the B cells with C57BL/6 IL-10R1 was decreased. The level of CD16/32 on the B cells with MRL IL-10R1 was lower than on the B cells with MRL IL-10R1 with the stimula- tion of IL-10. And the level of LECAM-1 on the former was higher than on the latter. Conclusion: IL-10R1 of MRL mouse loses the inhibition ability of proliferation, activation and tendency migration to B lymphocytes, which is maybe associated with the occurrence and development of SLE.
