摘要
目的:为了观察藜芦酸在LPS刺激的RAW264.7细胞中对一氧化氮(NO)及前炎性因子等表达的抑制作用。方法:采用Griess试剂对RAW264.7细胞培养上清中的NO进行检测;用ELISA法对IL-6、IL-1β及TNF-!等前炎性因子进行分析;并通过Western blot法对诱导型NO合酶(iNOS)、STAT-1及NF-"B的表达进行检测。结果:藜芦酸能显著抑制由LPS刺激的RAW264.7细胞中的NO产生及iNOS的表达,并呈剂量依赖的特点;且还检测到藜芦酸能抑制IL-6的产生,此外,藜芦酸还能抑制STAT-3及NF-"B的磷酸化。结论:本研究揭示了藜芦酸可以中和炎症疾病中产生的过量的NO。
Objective:To evaluate the inhibitory effect of veratric acid on nitric oxide (NO) production and pro-inflammatory cytokines in LPS-stimulated RAW264.7 ceils. Methods: NO production of RAW264.7 cells induced by veratric acid was determined by Griess reagent; IL-6, IL-1β and TNF-a cytokines were tested by ELISA; and the expression of inducible nitric oxide synthase (iN- OS), STAT-1 and NF-kB were measured by Western blot. Results: It was found that veratric acid inhibited NO production and induc- ible nitric oxide synthase (iNOS) expression in LPS-stimulated RAW264.7 cells and in dose-dependent manner. The inhibitory effects of veratric acid on the generation of IL-6 was determined. Furthermore, veratric acid facilitated the inactivation of STAT-3 and NF-kB. Conclusion : These findings suggest that the treatment of veratric acid might be effective in neutralizing the over-expression of NO in in- flammatory disorders.
