强骨康疏胶囊对去卵巢骨质疏松症模型大鼠细胞因子的影响 被引量:11

摘要 目的:通过观察去卵巢骨质疏松症模型大鼠骨密度以及血清BALP、TRACP、MMP-2、TGF-β1的变化,研究强骨康疏胶囊治疗去卵巢骨质疏松症模型大鼠的作用机制。方法:采用40只6月龄SD大鼠,随机分为空白对照组、模型对照组、雌激素组、中高组以及中低组。采用去除大鼠双侧卵巢的方法复制骨质疏松症模型。各组大鼠灌胃1月后,心脏取血法采得血液并分离血清,并取右侧股骨行骨密度检测,通过测量大鼠股骨骨密度以及检测血清BALP、TRACP、MMP-2、TGF-β1含量来评定造模是否成功以及强骨康疏胶囊对骨质疏松症模型大鼠细胞因子的影响。结果:中高组能明显提高骨质疏松症模型大鼠骨密度(P<0.05);与模型对照组相比,中高组能显著增加反映骨形成的血清学指标BALP的含量(P<0.001),降低反映骨吸收水平的血清TRACP水平(P<0.05)以及反映骨转换的血清MMP-2含量(P<0.05),还能刺激骨TGF-β1的分泌(P<0.05)。结论:强骨康疏胶囊能提高去卵巢骨质疏松模型大鼠的骨密度;对骨质疏松症大鼠的骨代谢有正向调节作用。强骨康疏胶囊对去卵巢骨质疏松症模型大鼠细胞因子的调节作用可能是其治疗原发性骨质疏松症的作用机制之一。 Objective:To evaluate the mechanism of Qiang Gu Kang Shu capsule (QGKS) in treatment of osteoporosis (OP) by observation of serum BALP, TRACP, MMP-2, TGF-β1 and bone mineral density (BMD) in ovariectomized osteoporosis rats. Methods: In this experiment, forty sprague dawley(SD) rats at age of six months were used, and were randomly divided into blank group, model group, estrogen group, high dose of traditional Chinese medicine group, as well as low dose of traditional Chinese medi- cine group. Excepted of the blank group, the method of removing both ovaries in the other groups' rats were used to copy osteoporosis model. After one month' s intragastric administration, blood was collected by the method of heart blood sampling, and then separated serum from it which was used to detection serum BALP,TRACP, MMP-2, TGF-β1. The right femur was separated for BMD testing that also can be used by justify whether the OP model were correctly copied. Results: Compared to the model group, high dose of QGKS could increase the OP rats' BMD( P 〈 O. 05), and eould obviously increase BALP ( P 〈 0.001 ) in serum which was the sign of bone information, it couhl reduce the serum TRACP and MMP-2 in OP rats, that were the signs of bone resorption, also, it could stimulate synthetizing serum TGF-β1 in OP rats. Conclusion: This study proves that QGKS has the effects of promoting the BMD in ovarieeto- mized OP rats. The effect of QGKS on cytokines in ovariectomized OP rats may be one of the mechanisms that how it played a good therapeutic effect on primary osteoporosis.
