摘要
目的:对比观察益肾蠲痹丸对肾虚证与脾虚证胶原诱导性关节炎(CIA)大鼠的治疗作用。方法:选用6周龄雌性SD大鼠114只,随机分为10组,分别采用去势法和羟基脲法制作肾虚模型4组、利血平法制作脾虚模型2组,在此基础上制作CIA模型。免疫18d后,4个治疗组均灌胃给予益肾蠲痹丸。给药4周后检测各组大鼠关节破坏程度及肾虚与脾虚状态。结果:(1)益肾蠲痹丸可以显著减轻肾虚证CIA大鼠膝关节骨破坏,对去势肾虚大鼠的关节保护作用更显著。(2)益肾蠲痹丸可显著升高去势肾虚大鼠血浆中血浆皮质酮含量及肾上腺cAMP/cGMP;益肾蠲痹丸能显著降低脾虚证CIA大鼠T细胞增殖能力。结论:益肾蠲痹丸对肾虚证、脾虚证及未施加证候处理因素的CIA大鼠均具有治疗作用,对肾虚证CIA大鼠的治疗作用要好于脾虚证及未施加证候处理因素的CIA大鼠。去势法制作的肾虚证CIA模型更符合中医的肾虚证候动物模型。
Objective:To evaluate the effect of Yi Shen Juan Bi Pill (YJB)on TCM kidney deficiency and spleen deficiency in collagen induced arthritis (CIA)rats.Methods:A total of 114 six-week-old female Sprague-Dawley rats were randomly divided into 10 groups,castration surgery and hydroxyurea was used to built kidney deficiency and spleen deficiency model respectively.Then cattle ]] type collagen was used to make arthritis model (except for the rats of the normal control group).YJB was used to treat CIA rats with TCM kidney deficiency or spleen deficiency from the 18th day after immunity for 4 weeks.The degree of joint damage and syndromes were used to evaluate the effect of YJB.Results: (1)YJB treatment can prevent the CIA rats form joint damage,which was more effective in CIA rats with kidney deficiency than CIA rats and CIA with spleen deficiency rats,and more effective in castrated CIA rats than hydroxynrea CIA ones.(2)YJB improved the ievel of corticosterone and cAMP/cGMP in castrated CIA rats and also decreased the prolifera- tion of T cells in spleen deficiency CIA rats.Conclusion:The protective effect of YJB can been seen on all the CIA rats.It is more effective in CIA rats with kidney deficiency than the other two models,and more ef- fective in castrated CIA rats than hydroxyurea CIA ones.The kidney deficiency syndrome produced by castra- tion in CIA model is more in line with TCM kidney syndrome in animal models.
