藠头组培快繁技术研究

Studies on the rapid propagation of Allium chinense G. Don by tissue culture

摘要 以藠头(Allium chinense G.Don)鳞茎为外植体,采用MS培养基进行组织培养,对培养基中附加的不同浓度6-BA、NAA和活性炭对藠头不定芽的诱导率及诱导数目的影响进行了正交实验分析,并对光照条件对藠头不定芽诱导的影响进行了单因素LSD检验。结果表明,藠头不定芽诱导的最佳培养基为MS+1.5 mg/L 6-BA+0.1mg/L NAA+1.0 g/L活性炭,诱导率可达90%以上,芽平均数为2.0以上,且状态良好。光照条件对藠头不定芽的诱导率有显著的影响,充足的光照对芽的生长有促进作用,而培养的最初48h,应采取暗培养,之后将其转到光照8小时/黑暗16 h下培养,大约7 d后,将培养瓶转到光照16 h/黑暗8 h下培养,诱导出的芽生长旺盛。 With the bulbs of Allium chinense G. Don as explants, MS medium was used to the tissue culture. The influence of different concentration of 6 - BA, NAA and activated carbon in the medium on the ratio of adventitious - bud induction and the average number of buds was analyzed through orthogonal design method. The influence of light condition on the ratio of buds induction was tested by LSD. The results showed that the optimum formula of medium for adventitious buds induction was MS + 1.5 mg/L 6 - BA + 0.1 mg/L NAA + 1.0 g/L activated car- bon, the ratio of induction was more than 90%, the average number of buds was above 2.0, and the quality of buds was in good condition. Light conditions had a significant impact on the ratio of adventitious bud induction of A. chinense. Sufficient light could stimulate the growth of buds. At the first 48 hours, the culture should be taken in dark, then was transferred in 8 hours of light/16 hours of dark, and about a week later, in 16 hours of light/ 8 hours of dark. In this light condition, the induced buds grew strong.
